P-2: Impact of Sesame Oil Supplementation onDiabetic Rat Epididymal Sperm
Authors
Abstract:
Background: Formation of hydrogen peroxide derivativesdue to metabolic disturbances in the male diabetics can affect their fertility. The antioxidant property of sesame oil has been documented, previously. The primary aim of the present study was to find if sesame oil supplementation can reduces the impact of diabetes on sperm parameters in rat or not. Materials and Methods: Eighteen numbers of male Wistar rats assigned to four experimental groups: Control (n=3); intact animals received no treatment, Diabetic rats (n=5); diabetic rats treated with 5 (n=5) and 10% (n=5) of sesame oil within their diet throughout 60 days of experiment. Diabetes induced by IV injection of 45 mg/kg streptozotocin dissolved in citrate buffer (PH= 4.5). After 60 days of experiment, the animals were euthanized and their testes-epididymis dissected and weighted by digital balance. Left cauda epidydimis were separated and total recovered sperms during 4 hrs of incubation in the normal saline (35- 37°C) were counted and divided by the weight of left testis and left epidydimis to find total sperm/g testis and epidydimis, respectively. Contra-lateral cauda epidydimis dissected from testis and a sample of sperm isolated in normal saline (35-37°C). Sperm progressive motility (SPM) of recovered sperms was analyzed subjectively with light microscopy and analyzing four different fields of slides. Sperm viability was analyzed with conventional stain Eosin B/nigrosin stain. Data analyzed by ANOVA and expressed as Lsmeans ± ESM.Results: Diabetes (228 ± 19.04 g) severely (p0.05). There was no significant difference between groups according to testes and epidydimis weights (p>0.05). Surprisingly, total sperm/g testis and epidydimis were not affected by diabetes (28.5 × 106) (p>0.05) compare to control (28.7 × 106) while they were severely influenced by sesame oil supplementation, either 5 (19.3×106) or 10 (14.1×106) %. Diabetes (19.4 ± 2.9) and 10 % supplementation of sesame oil (21 ± 2.9) severely affected (p

P-85: Epididymal Sperm Parameters following Sperm Mediated Gene Transfer of EDFP-pIRES Vector in Rat
Background: Sperm mediated gene transfer was approved for producing transgenic animals. There are many studies which reported different success rate by this procedure. Failure in this project mostly is returned to transfection and stability of transferred genes. The other problem that may contribute in project failure may return to the testicular damages which occur at the time of transfection,...
full textP-2: Evaluation of Epididymal Sperm Quality, DNA Damage and Sperm Maturation Abnormality in Streptozotocin- induced Diabetic Mice
Background: Diabetes mellitus, a condition of chronic hyperglycemia, represents one of the greatest concerns to modern global health. Diabetes is a serious metabolic disorders with numerous complications. It is well known that, elevation of blood glucose levels leads to structural and functional changes in various target tissues and organs. The objective of the present study was to verify if th...
full textEffects of Sesame Oil on the Reproductive Parameters of Diabetes Mellitus-Induced Male Rats
PURPOSE The purpose of the present study was to investigate the effect of sesame oil on the reproductive parameters of diabetic male Wistar rats. MATERIALS AND METHODS The adult male rats in a split plot design were divided into normal (n=10), normal 5% (n=5; 5% sesame oil enriched diet), diabetic (Streptozocin induced diabetes; n=9), diabetic 5% (n=9; 5% sesame oil enriched diet), and diabet...
full textThe Effect of Chronic Spinal Cord Injury on Ultrastructure of Epididymal Sperm in Rat
Purpose: The effects of chronic SCI on ultrastructure of spermatozoa aspirated from epididymis of rats. Materials and Methods: 45 adults Wistar rats were divided into 3 groups of SCI, control and sham. Following laminectomy, SCI was induced with a 15g weight dropped from a distance of 10 cm, onto exposed dura matter at T10 level. Sham group underwent laminectomy of T10 only, while control was n...

Background: Motile sperm is essential for successful embryo production. The aim of the present study was to compare the in vitro embryo development following isolation of Ram epididymal sperm isolation by swim up (SP) or Histoprep® continuous gradient (HP). Materials and Methods: Ram testes-epididymis were provided from the local slaughterhouse and transported to the lab. Sperms were recovered ...

Background: Methylphenidate is one of the most common medications used for treatment of attention-deficit/hyperactivity disorder. It may be used for maintaining alertness and improving of attention which may lead to increase of the risk of substance abuse in some cases. The occurrence of alteration in male reproductive system such as hormonal secretion changes and alteration of testicular sperm...
